A recently granted patent (Publication Number: US11946063B2) discloses a transgenic mouse with a unique genetic makeup aimed at enhancing the expression of immunoglobulin heavy chain variable regions. The mouse’s genome includes a combination of human variable region coding sequences and mouse non-coding sequences, along with a long DH (LDH) expression cassette that replaces all endogenous DH segments. This expression cassette contains 5-60 different human DH gene segments, with at least two directly fused segments encoding a heavy chain CDR3 (HCDR3) amino acid sequence of at least 10 amino acids. The fused DH gene segments consist of a mutated human DH gene segment devoid of stop codons and a human DH gene segment flanked by a mouse intergenic region, enabling the expression of a long HCDR3 amino acid sequence of at least 20 amino acids.

Furthermore, the patent details additional features of the transgenic mouse, such as the length of the HCDR3 amino acid sequence ranging from 20-61 amino acids, the connection of fused DH gene segments by a mouse intergenic region, and the inclusion of bovine DH gene segments in the recombinant DH construct. Additionally, the construct may contain human DH gene segments with deleted stop codons or fused with mouse intergenic regions, further enhancing the expression of the long HCDR3 amino acid sequence. These unique genetic modifications in the transgenic mouse aim to improve the production and functionality of immunoglobulin heavy chain variable regions, potentially leading to advancements in research and development in the field of immunology and antibody engineering.
